In-vitro-Diagnostik

Der Schwerpunkt der Arbeitsgruppe „Zelluläre allergologische Diagnostik“ liegt auf dem Nachweis der Aktivierung von basophilen Granulozyten nach Allergenstimulation mittels Flowzytometrie (Basophilenaktivierungstest – zellulärer Allergenstimulationstest für Typ-I-Allergien). Mit dieser Methodik können patientenbezogene diagnostische Fragestellungen vor allem im Bereich der Insektengiftallergie und bei Nahrungsmittelallergien beantwortet werden. Darüber hinaus ist dieser zelluläre Test seit Jahren für wissenschaftliche Forschungsansätze im Bereich IgE-vermittelter Reaktionen an unserer Klinik etabliert. Hier besteht eine Zusammenarbeit mit dem Zentrum Allergie und Umwelt (ZAUM), TUM und Helmholtz Zentrum München „Translational Biochemistry group“ (PD Dr. S. Blank) und dem Department of Infection and Immunity des Luxembourg Institute of Health (Prof. M. Ollert).

Darüber hinaus wird derzeit der Lymphozytentransformationstest (zellulärer Allergenstimulationstest für Typ-IV-Allergien) etabliert, bei dem aus dem peripheren Blut isolierte Lymphozyten und Monozyten in vitro mehrere Tage in Gegenwart von Allergenen kultiviert werden. Die einsetzende Proliferationsantwort der das Antigen erkennenden Lymphozyten wird als Ausdruck der Sensibilisierung gemessen. Der Lymphozytentransformationstest kann zur Diagnostik bei Arzneimittelreaktionen und Kontaktallergien sowie für wissenschaftliche Fragestellungen eingesetzt werden.
